There's a very low chance he's not already aware of it. Personally, I feel uncomfortable when someone points out or gives me advice about my stutter. I struggle with confidence and it makes me feel like my speech must be really bothering them if they feel the need to say something. Your coworker might have already done speech therapy when he was younger and worked hard to improve his speech, so any comment could feel like a setback to him.
